NUCLEOSIL®, a classical silica manufactured by MACHEREY-NAGEL GmbH & Co. KG is available in a wide range of pore sizes. NUCLEOSIL 100Å and 120Å have similar relative retention characteristics. NUCLEOSIL 100Å has the greater capacity whilst the 120Å material exhibits the lower column back pressure. The wide pore NUCLEOSIL 300Å silica is used as the base material for the reversed-phase bioanalytical columns.NUCLEOSIL 100-5C18AB was the first of the new generation NUCLEOSIL materials which include the HD high density phases NUCLEOSIL Protect 1 and the Nautilus, for use with up to 100% aqueous eluents.

NUCLEOSIL® Base Deactivated Phases


NUCLEOSIL® 100-5 C18 AB, NUCLEOSIL® 100-5 C8/C18 HD and NUCLEOSIL® 100-5 Protect 1 represent three successive generations of base deactivated reversed phase HPLC columns, manufactured from purer silica. NUCLEOSIL® 100-5 C18 HD features a high-density mono-layer coverage. Due to the shorter chain and anincorporated group,  NUCLEOSIL®100-5 Protect 1 exhibits a distinctive polar selectivity. The HD material is also offered in an octyl-bonded chemistry. Figure 1 compares the selectivity differences of four NUCLEOSIL® base deactivated phases. For the Protect 1 phase, elution of naphthalene and ethylbenzene is reversed.

NUCLEOSIL® C18 Nautilus


NUCLEOSIL® C18 Nautilus shows outstanding stability in highly aqueous media (>95%) due to a covalently bonded C18 silane with an embedded polar functional group. As well as the hydrophobic interactions between analyte and stationary phase shown by standard C18 phases, NUCLEOSIL C18 Nautilus also shows polar interaction via hydrogen bonding and dipole-dipole forces. This can influence retention time and improve selectivity, particularly for the separation of polar compounds
